A CAR mainly consists of a single chain variable fragment (scFv) from a monoclonal antibody which binds a specific antigen, a hinge and spacer element, a trans-membrane region and an internal signaling domain such as CD3ζ. The main advantage of CAR-T cell is that it bypasses MHC restriction, which allows for direct activation of effector cells for the treatment of various types of tumors.
