Due to the low abundance of membrane proteins in their natural hosts, it is necessary to overexpress these proteins in heterologous systems. Conventional cell-based systems can be problematic in insufficient membrane insertion, precipitation of de novo synthesized proteins, or even cytotoxicity due to the extensive alterations in the host cell's metabolism.
